EXPRESSION OF FUNCTIONAL TOLL-LIKE RECEPTOR-2 AND -4 ON ALVEOLAR EPITHELIAL CELLS DURING HYPEROXIA-INDUCED OXIDATIVE STRESS

ABSTRACT
Background and aims: Expression of Functional Toll-like Receptor-2 and -4 on Alveolar Epithelial Cells during hyperoxia-induced Oxidative Stress.
Aims: To observe the dynamic changes and the effects of calcitonin gene-related peptide (CGRP) in the hyperoxia-induced lung injury
Methods: Sixty-four SD Rat were divided randomly into eight group(8 animals each),Four groups were exposed to oxygen for 1,3,7,and 14 days, respectively. The control group breathed only filtered room air, the subjects was sacrificed. The changes of CGRP andα-CGRPmRNA andβ-CGRPmRNA expression were measured by radio-immunity technique and RT-PCR. The changes of lung histomorphology was observed.
Results: In the model group compared with those of the air control,the concentration of CGRP increased significantly on 3 days, then returned to the normal level at day 14; The changing pattern of the CGRP mRNA in the lung weren’t similar to those observed for CGRP, α-CGRPmRNA was expression increased significantly on 1 day to 7 days,butβ-CGRPmRNA on 3 days to 14 days; The histopathological examination demonstrated different degrees of alveolitis, broaden interstitium and reduced alveolar quantity in the Model group compared with control group.
Conclusions: CGRP may play an important role in the development of hyperoxia-induced lung injury. bothα-CGRP mRNA andβ-CGRP mRNA were expressed in hyperoxia-induced lung injury.
